The protein possesses four predicted transmembrane domains and intracellular amino and carboxyl-terminal domains. The ORAI1 gene is localized on human chromosome 12q24.31.

ORAI calcium release-activated calcium modulator 1 (ORAI1) modulates Ca²⁺ entry into cells. The channel is activated by stromal interaction molecule-1 (STIM-1). Mutation in the gene encoding the protein has been linked to late-onset tubular aggregate myopathy and congenital miosis.
