BackgroundA novel member of the poly(ADP-ribose) polymerase (PARP) family, PARP10 has been characterized as a novel c-myc-interacting protein. PARP family members regulate transcription, genomic stability, and DNA metabolism, especially in response to DNA damage. PARP10 can autopolyribosylate as well as ribosylate core histones and is found in both the cytoplasm and nucleus. In cells, PARP10 associated with the nucleolus is phosphorylated in a Cdk dependent manner. In vitro Cdk2/cyclin E can regulate PARP10 activity through phosphorylation. Phosphoryated PARP-10 is absent in growth-arrested cells. Overexpression of PARP10 results in loss of cell viability, while down-regulation results in delayed G1 progression and apoptosis. These results suggest that PARP10 functions in cell proliferation and may serve as a marker for proliferating cells.
