General description

Chk2 is a protein kinase activated and phosphorylated in response to DNA damage, especially DNA double-strand breaks. As a cell cycle checkpoint regulator it acts to prevent DNA replication and inhibit mitotic entry prior to the conclusion of DNA replication. In response to DNA damage, the checkpoint kinase ATM phosphorylates and activates Chk2, which in turn directly phosphorylates and activates p53. Chk2 also phosphorylates and activates BRCA1, the product of a tumor suppressor gene mutated in breast and ovarian cancer.
There are 12 known isoforms of Chk2. The majority of which are expressed solely in the cell nucleus. The exception is isoform 10 which is found throughout the cell. High expression has been observed in spleen, peripheral blood leukocytes, colon and testis. Lower expression is observed in other tissues. Mutations in CHEK2 gene coding for Chk2 are associated with Li-Fraumeni syndrome 2, and have been observed in some patients that contracted prostate cancer and osteosarcoma.
The observed molecular weight is 61 kDa UniProtKB/Swiss-Prot O96017 (CHK2_HUMAN) describes 12 isoforms produced by alternative splicing.
