General description

Eukaryotic initiation factor 4A-II, (EIF4A2), or ATP-dependent RNA helicase eIF4A-2 is the critical RNA helicase protein required for mRNA cap recognition and mRNA binding to the ribosome. EIF4A2 is part of a multi-subunit complex that allows translation of mRNAs to begin. EIF4A2 unwinds the RNA secondary structure in the 5’-UTR region and thus allows the small ribosomal subunit to bind the mRNA and begin scanning for the initiator codon. EIF4A2 also plays a key role in mRNA expression inhibition. Recent research demonstrates that inhibition of EIF4A2 function via microRNA inhibition is required first, which then leads to mRNA destabilization and ultimately translation suppression. EIF4A2 is expressed in all tissues, as well as in cancerous tumors. Expression is very high in ovary, followed by liver, kidney, brain and pancreas in lower amounts. EIF4A2 is often used as a “housekeeping” marker as well because of its wide expression.

Immunofluorescence Analysis: A 1:25 dilution from a representative lot detected EIF4A2 in HepG2 cells.

Preparation Note

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.

Analysis Note

Evaluated by Western Blotting in MCF7 cell lysate.

Western Blotting Analysis: A 1:500 dilution of this antibody detected EIF4A2 in MCF7 cell lysate.
