Phospholipase C (PLC) is a group of enzymes that play a critical role in signal transduction during the hydrolysis of membrane phospholipids as a response to extracellular stimuli. Part of the phosphinositide-specific PLC family, PLC У-1 is known for its involvement in growth-factor-dependent signal transduction. PLC У-1 is made up of 2 SH2 domains, one SH3 domain and 2 tyrosine phophrylation sites. Almost all growth factors can stimulate tyrosine phosphorylation and subsequent activation of PLC У-1. (Tvorogov, 2005). Of the protein kinases that stimulate tyrosine phosphorylation, PLC У-1 is mostly dependent on three different kinds; ZAP-10, Lck, and Itk. (Kang, 2003). Activation of PLC У-1 catalyzes the hydrolysis of phosphatidylinositol 4,5-biphosphate which in turn produces two second messengers, diacylglycerol (DAG) and inositol 1,4,5-triphosphate(IP3). DAG serves to activate protein Kinase 3, while IP3 mediates the release of calcium. PLC У-1 is crucial in its role to the mediation of cellular proliferation and differentiation.
