General description

This product has been discontinued.

Recognizes the ~250-260 kDa IGF-1 receptor protein in MCF-7 cells. Does not cross-react with recombinant human IGF-I binding protein 1, IGF-I, or IGF-II.

Protein G purified mouse monoclonal antibody. Recognizes the ~250-260 kDa IGF-1 receptor protein.

Immunogen

Epitope: Extracellular domain of IGF-I Receptor
recombinant, human, soluble IGF-I receptor

Application

ELISA (2 µg/ml, capture antibody)

Immunoblotting (non-denaturing protocols only)

Neutralization Studies (11 µg/ml, see comments)

Physical form

Lyophilized from PBS, 20 mM ammonium bicarbonate, ≤5% trehalose, 100 µg BSA.

Preparation Note

Reconstitute with sterile PBS to the desired concentration. Following reconstitution, antibody can be stored at 4°C for up to 1 month. For long-term storage, aliquot and freeze (-20°C).

Analysis Note

Positive Control
Recombinant IGF-I receptor protein or MCF7 cells

Other Notes

Endotoxins: ≤1 EU/µg anti-IGF-IR. This antibody was selected for its ability to block cell surface IGF-I receptor mediated IGF-I and IGF-II bioactivities. The exact concentration of antibody required to neutralize the human cell surface IGF-I receptor mediated bioactivity is dependent on the IGF concentration as well as on the number and types of IGF receptors present on the cell surface (as a function of cell type and culture conditions). A concentration of 11 µg/ml neutralizes ~50-75% of IGF-I receptor-mediated rhIGF-I biological activity in MCF7 cells. This antibody recognizes a conformational epitope. The detection limit for human recombinant IGF-I receptor by immunoblotting is approximately 25 ng/lane. The detection limit for human recombinant IGF-I receptor is ~0.16 ng/well. Based on immunoblots and ELISA, this antibody does not cross-react with human recombinant IGF-I bp1, human recombinant IGF-I and human recombinant IGF-II. Antibody should be titrated for optimal results in individual systems.
