General description

Angiopoietin-1 (Ang1) and its naturally occurring inhibitor, angiopoietin-2 (Ang2) exert their effects by inducing autophosphorylation of their signaling receptor Tie2 (also known as Tek). Phosphorylation of the receptor occurs during vasculogenesis and is required for its activity in regulating endothelial cell proliferation, differentiation and guiding the proper patterning of endothelial cells during blood vessel formation. Tie-2 is a 140 kDa transmembrane receptor-tyrosine kinase that is expressed by endothelial cells and primitive hematopoietic tissues. Tie2/Tek has been shown to be critical for endothelial cell survival, which is evidenced in part by the constitutive phosphorylation that is observed in quiescent endothelium. More recently a multifunction docking site has been identified on the receptor (Tyr1100) that serves as a binding site for other partners including Grb2, Grb7, and p85 and acting via an SH2 domain interaction.
Rabbit polyclonal antibody supplied as undiluted serum. Recognizes the ~140-145 Tie2 protein phosphorylated at Tyr1102 and Tyr1108.
Recognizes the ~140-145 Tie2 protein phosphorylated at Tyr1102 and Tyr1108 in colon carcinoma tissue.
This PhosphoDetect Anti-Tie2 (Ab-1) (pTyr1102/1108) Rabbit pAb is validated for use in ELISA, Paraffin Sections for the detection of Tie2 (Ab-1) (pTyr1102/1108).

Immunogen

a synthetic phosphopeptide [EERRTpYVNTTLpYEK(C)] corresponding to amino acids 1097-1110 of human Tie2, where the K in position 1100 of the native sequence has been substituted with R

Application

ELISA (see comments)
Paraffin Sections (1:2000-1:5000, heat pre-treatment required)

Physical form

Undiluted serum.

Preparation Note

Following initial thaw, aliquot and freeze (-20°C).

Analysis Note

Positive Control
Colon carcinoma tissue

Other Notes

Can be used in conjunction with a biotinylated phosphopeptide to measure the level of Tie2 activity by competition ELISA. Antibody should be titrated for optimal results in individual system.
