An N-terminally modified human Aβ1-40 peptide labeled with a fluorophore, 7-diethylaminocoumarin-3-carbonyl (DAC) that shares a common binding site with Aβ1-40 (Cat. No. 171590) with 40-fold greater affinity. Displays stronger affinity towards GM1-ganglioside and cholesterol containing membranes but little affinity for phospholipids. Exhibits only a weak fluorescence in aqueous medium; however, it strongly fluoresces upon binding to membranes. Useful in assessing the binding affinity of Aβ for various membranes.

      Revision11-April-2008 RFH
      SynonymsDAC-Aβ1-40, Human
DescriptionAn N-terminally modified human Aβ1-40 peptide labeled with a fluorophore, 7-diethylaminocoumarin-3-carbonyl (DAC), which shares a common binding site with Aβ1-40 (Cat. No. 171590). However, it displays about 40-fold greater affinity than Aβ1-40. Exhibits only a weak fluorescence in aqueous medium; however, it fluoresces strongly upon binding to the membranes. Displays greater affinity towards the GM1-ganglioside and cholesterol-containing membranes but little affinity for phospholipids. A reduction in GM1 content is reported to reduce DAC-Aβ1-40 binding to membranes. DAC fluorescence can be used to assess the binding affinity of Aβ for various membranes. Excitation max.: ~430 nm; emission max.: ~470 nm.
