      Application CommentsThis antibody has been selected for its ability to neutralize the biological activity of human recombinant angiogenin. The activity of human recombinant angiogenin was measured based upon its ribonucleolytic activity toward yeast tRNA. Using the assay conditions described, 1 µg human recombinant angiogenin produces an absorbance change at 260 nm of ~2.0-3.0. The suggested antibody neutralization concentration required to yield 50% of the activity due to 1 µg of human recombinant angiogenin is approximately 100-150 µg of the antibody. The detection limit for human recombinant angiogenin by immunoblotting is ~5 ng/lane under reducing and non-reducing conditions. The detection limit for human recombinant angiogenin by ELISA is ~0.3 ng/well. This antibody exhibits no cross-reactivity with other cytokines when tested in ELISA. Antibody should be titrated for optimal results in individual systems.

      DescriptionImmunoaffinity purified goat polyclonal antibody. Recognizes the ~14 kDa angiogenin protein.
      BackgroundAngiogenin was initially isolated from culture medium conditioned by human colon adenocarcinoma cells (HT-29 cells) as a ~14 kDa protein homologous to pancreatic ribonuclease A. Angiogenin is a potent inducer of blood vessel formation whose enzymatic and three-dimensional relationship to RNAse A is critical for its biological function. It exerts ribonucleolytic activity that is several magnitudes lower than that of RNAse A but nonetheless essential for angiogenesis since inhibition of RNAse activity will also abolish angiogenesis. Internalization and translocation to the nucleolus are also required for activity. Angiogenin binds to high affinity endothelial cell receptors and with lower affinity to extracellular matrix components. An angiogenin binding protein (~42 kDa) previously isolated from the cell surfaces of endothelial cells has been shown to be a member of the actin family by peptide mapping and partial amino acid sequencing. It has also been shown in vitro that angiogenin absorbed on plastic will support the cell adhesion of both endothelial cells and fibroblasts. Angiogenin is differentially produced by anchorage-dependent growing cells including vascular endothelial cells, aortic smooth muscle cells, fibroblasts and tumor cells.
