Background InformationPACT is also known as a protein activator of the interferon-induced protein kinase, PKR. PACT is a heterodimer with 3 double stranded RNA binding domains (DRBM) and acts as an antiviral mediator, growth regulator and signal transducer stimulated by stress. PACT activates EIF2AK2 in the absence of double stranded RNA binding via the DRBM domains. Suppression of PACT occurs in the presence of growth factor and is thought to be involved in dystonia type 16, which leads to axial muscle movement, Parkinsonian features and sardonic smile.

      Entrez Gene Number
Entrez Gene SummaryThis gene encodes a protein kinase activated by double-stranded RNA which mediates the effects of interferon in response to viral infection. Mutations in this gene have been associated with dystonia. Alternative splicing results in multiple transcript variants. [provided by RefSeq].

      UniProt Number
UniProt SummaryFUNCTION: Appears to have a pro-apoptotic function that may be suppressed in the presence of growth factor By similarity. Activates EIF2AK2 in absence of double stranded RNA (dsRNA).

SUBUNIT: Heterodimer. Interacts with EIF2AK2 through its DRBM domains. Also able to interact with dsRNA. Ref.1

PTM: Phosphorylated on serine By similarity.

DISEASE: Defects in PRKRA are the cause of dystonia type 16 (DYT16) [MIM:612067]. Affected individuals have progressive, generalised, early-onset dystonia with axial muscle involvement, oromandibular (sardonic smile), laryngeal dystonia and, in some cases, parkinsonian features. Ref.14 Ref.15

      SIMILARITY: Contains 3 DRBM (double-stranded RNA-binding) domains.

      Molecular Weight~34 kDa.
