      An aqueous-based mounting medium designed for the permanent preservation of alkaline phosphatase and immunoperoxidase stained cells and tissue sections. ChromoSave™ Reagent can be used with organic solvent soluble horseradish peroxidase chromogens, such as 3-amino-9-ethylcarbazole (AEC) and 4-chloro-1-napthol, as well as alkaline phosphatase chromogens, such as 5-bromo-4-chloro-3-indolyl phosphate/nitroblue tetrazolium (BCIP/NBT) and Naphthol AS-MX phosphate/Fast Red TR salt. It can also be used with tissue sections stained with chromogens that are insoluble in organic solvents, such as diaminobenzidine (DAB), eliminating the dehydration step. ChromoSave™ Reagent is also compatible with fluorescein, rhodamine, and TEXAS RED® stained tissues. For preservation of phycoerythrin, phycocyanin, or allophycocyanin stained tissue samples, use FluorSave™ Reagent (Cat. No. 345789).

      DescriptionAn aqueous mounting medium designed for the permanent preservation of peroxidase-and alkaline phosphatase-stained tissue sections. Hardened ChromoSave™ forms an impervious barrier to organic solvent-based mounting media, such as Permount. When properly mounted with ChromoSave™, samples exhibit enhanced contrast and maintain their original clarity and strength of color. ChromoSave™ works well with organic solvent-soluble chromogens such as AEC, 4-chloro-1-naphthol, Naphthol AS-MX phosphate/Fast Red TR salt, and BCIP/NBT, as well as chromogens that are insoluble in organic solvents, such as DAB, thus eliminating the dehydration procedure. ChromoSave is also compatible with fluorescein-, rhodamine-, Texas Red-, and FluoroBlue-stained specimens. Not suitable for specimens stained with Phycoerythrin, Phycocyanin, or Allophycocyanin.

      Recommended reaction conditions
      Do not use oil directly on top of dried ChromoSave. Instructions for mounting with ChromoSave™ Reagent 1. Before using this product, remove the red cap covering the tip of the bottle and cut off the tip of the nozzle using a sharp blade or scissors. Re-cap the bottle of ChromoSave™ and store the bottle upside down in a cup or bottle. This eliminates the introduction of air bubbles into the medium. 2. Place all slides to be mounted on a horizontal surface and apply three drops of ChromoSave™ to the tissue sections. 3. Rotate the slides covered by the ChromoSave™ so that it spreads to cover an area roughly the size of a quarter. Do not apply a coverslip on top of the ChromoSave™. 4. Place the slides horizontally in an oven at 70-80°C for at least 10 min or 20-37°C for 2-3 h. Note: when using fluorescent dye-stained specimens, bake at 60-70°C for 20 min. 5. Remove the slides from the oven and allow them to reach room temperature. The slides are now ready for microscopic visualization. Note: microscopic air bubbles may be found after applying ChromoSave™ to particular tissue sections, especially gastrointestinal sections or necrotic areas on the sections. Slow drying of the ChromoSave™ at room temperature may help eliminate this occurrence. Post-Mounting ChromoSave™ Reagent with Permount ChromoSave™ is not suitable for use directly with oil. Slides with hardened ChromoSave™ can be post-mounted and cover-slipped in Permount if desired. Place 3 drops of Permount on top of the hardened ChromoSave™ and apply a coverslip. Allow the slides to dry at room temperature. The hardened polymer in ChromoSave™ has a refractive index very close to that of Permount. Do not heat slides after the application of Permount. Removal of ChromoSave™ Reagent When required, solidified ChromoSave™ may be partially or completely removed only if the slides have not been post-mounted with Permount. Immerse the slides in distilled water and very gently agitate the slides to assist in the removal of the ChromoSave™ layer. Repeat this procedure until the film of ChromoSave™ is removed. If overnight incubations are required, add sodium azide to a concentration of 0.05% to avoid bacterial growth. Excessive soaking in water may result in the detachment of the tissue sections from the glass slides. Following removal of the ChromoSave™, treat the tissue sections with a buffer compatible with the staining procedure to be used. Failure to re-stain the slides should not be considered as negative staining since incomplete removal of the ChromoSave™ may interfere with some staining procedures. Variables associated with assay conditions will dictate the proper working dilution.
