      CrmA (cowpox viral serpin cytokine response modifier A) is purified from E. coli transformed with a construct containing the full-length coding region of the CrmA gene and 7 additional amino acids that do not affect the activity. CrmA is a natural inhibitor of human caspase-1 and granzyme B, enzymes that are involved in apoptosis.

      DescriptionFull-length recombinant CrmA expressed in E. coli with 7 non-native amino acids (without affecting activity). Cowpox viral serpin cytokine response modifier A (CrmA) is a proteinase inhibitor that prevents programmed cell death initiated by a variety of stimuli. CrmA is selective in its ability to inhibit caspases, showing high affinity for interleukin-1β-converting enzyme (Caspase-1) and Granzyme B, enzymes that are involved in apoptosis.

      Recommended reaction conditionsFor highest inhibitory activity, activate CrmA with 2 mM DTT for 5 min at room temprature. If granzymeB room temperature. If granzymeB thiol ester-based substrate assays are to be preformed, the excess DTT can be blocked by adding 4 mM NaAsO2 to the activated CrmA.
