      DescriptionAnti-Bub1 Antibody, clone 14H5
Background InformationThe mitotic checkpoint prevents the resulting two daughter cells from having unequal numbers of chromosomes. This condition, known as aneuploidy, is present in almost all cancer cells and may be due to malfunctions in the mitotic checkpoint. Bub1 is a mitotic checkpoint kinase and has been implicated in the metaphase checkpoint control in mammalian cells.

      Applications
ApplicationDetect Bub1 with Anti-Bub1 Antibody, clone 14H5 (Mouse Monoclonal Antibody), that has been shown to work in IP, WB, ICC.
      Key Applications
      • Immunoprecipitation
      • Western Blotting
      • Immunocytochemistry
Application NotesWestern blot using an ECL detection system. Reacts with the ~150 kDa Bub1 protein.

Immunocytochemistry: 1 μg/mL on tissue culture cells. Suggested fix is 3.5% PBS-buffered paraformaldehyde for 7 minutes. Permeabilization method is 0.2% Triton-X-100 after fixation. Suggested blocking buffer is 10 mM Tris, pH 7.5 with 150 mM NaCl with 0.1% BSA.

Immunoprecipitation: 1 μg/mL. Preferred cell lysis buffer is RIPA. Final reaction volume is 200 μL with a final protein concentration in the reaction mix of 1 mg/mL. Suggested capture agent is Protein G.

Optimal working dilutions must be determined by the end user.

      Entrez Gene Number
Entrez Gene SummaryThis gene encodes a kinase involved in spindle checkpoint function. The kinase functions in part by phosphorylating a member of the miotic checkpoint complex and activating the spindle checkpoint. Mutations in this gene have been associated with aneuploidy and several forms of cancer.
      Gene Symbol
      • BUB1
      • hBUB1
      • BUB1A
      • BUB1L

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: O43683 # Involved in cell cycle checkpoint enforcement. Can interact and phosphorylate BUB3.
SIZE: 1085 amino acids; 122375 Da
SUBCELLULAR LOCATION: Nucleus (By similarity). Kinetochore (By similarity). Note=Nuclear in interphase cells. Kinetochore localization is required for normal mitotic timing and checkpoint response to spindle damage (By similarity).
TISSUE SPECIFICITY: High expression in testis and thymus, less in colon, spleen, lung and small intestine. Expressed in fetal thymus, bone marrow, heart, liver, spleen and thymus. Expression is associated with cells/tissues with a high mitotic index.
DOMAIN: SwissProt: O43683 CD1 domain directs kinetochore localization and binding to BUB3.
PTM: Phosphorylated upon DNA damage, probably by ATM or ATR.
DISEASE:SwissProt: O43683 # Defects in BUB1 are associated with tumor formation.
SIMILARITY: Belongs to the protein kinase superfamily. Ser/Thr protein kinase family. BUB1 subfamily. & Contains 1 CD1 domain. & Contains 1 protein kinase domain.
