      Background InformationMatrix protein 1 (UniProt: P05777; also known as M1 is encoded by the M gene in Influenza A virus. Matrix protein is the most abundant protein in virion that when expressed alone can form virus-like particles in transfected cells. It can exist as homodimer and homomultimer. It plays a critical role in virus replication, from virus entry and uncoating to assembly and budding of the virus particle. The influenza A virus M1 protein is shown to have lipid binding properties and interacts tightly with the plasma membrane. It forms a coat inside the viral envelope and acts as a bifunctional membrane/RNA-binding protein that mediates the encapsidation of RNA-nucleoprotein cores into the membrane envelope. M1 protein consists of two domains connected by a linker sequence. The N-terminal domain has a multi-helical structure that can be divided into two subdomains. The C-terminal domain also contains alpha-helical structure. Binding of M1 protein to ribonucleocapsids (RNPs) in the nucleus seems to inhibit viral transcription. Interaction of viral nuclear export protein (NEP) with M1-RNP is thought to promote nuclear export of the complex, which is targeted to the virion assembly site at the apical plasma membrane in polarized epithelial cells. Two isoforms of influenza virus A M1 protein have been described that are produced by alternative splicing. (Ref.: Gomes-Puertas, P., et al. (2000). J. Virol. 74(24); 11538-11547).
