General description
カルパインは、活性のためにカルシウムが不可欠な非リソソームシステインプロテアーゼです。主なカルパインアイソザイムには次の2種類があります：μ-カルパインおよびm-カルパイン。 in-vitroアッセイでは、活性のためにそれぞれマイクロモルまたはミリモルのカルシウムを要します。主なカルパインアイソザイムは2つのサブユニット、つまり80 kDa触媒サブユニットと30 kDa調節サブユニットで構成されます。この大・小のサブユニットは、チモーゲンのタンパク質分解活性後にカルシウム存在下で結合した状態を維持します。

Application
この抗カルパイン抗体、μ-またはm-カルパイン（カルパインIまたはII）の小サブユニット、クローンP1は、ELISA、WB、IC、IHを用いたカルパインの検出がバリデーションされています。
イムノブロッティング：1：500-1,000。μ-カルパインとm-カルパインの小サブユニットは、いずれも30 kDaです。ELISA：1：50,000（免疫原に対して）。免疫組織染色：1：100を4%パラホルムアルデヒドまたは1%パラホルムアルデヒド／4%グルタルアルデヒド固定組織で使用。免疫細胞染色：1：100。最適な希釈濃度は、ご自身で決定してください。

ヒト赤血球μ-カルパイン（カルパインI）またはウシ心筋m-カルパイン（カルパインII）の小サブユニットと反応します。
