General description
Glycoprotein VI (GPVI), or glycoprotein 6 (GP6), is a member of the immunoglobulin gene superfamily, closely related to Fc receptor gamma chain (FcRgamma) and natural killer receptors. GPVI is a collagen receptor involved in collagen-induced platelet adhesion and activation, and may contribute to arterial and venous thrombus formation. GPVI is a membrane glycoprotein that plays a crucial role in platelet procoagulant activity and subsequent thrombin and fibrin formation. GPVI is expressed in megakaryocytes and platelets. The signaling pathway of GPVI involves the FcRgamma, the Src kinases (likely Fyn/Lyn), the adapter protein LAT and leads to the activation of phospholipase C gamma2. It has been shown that bleeding disorders such as Bleeding Disorder Platelet-Type 11 (BDPLT11), a mild to moderate bleeding disorder caused by defective platelet activation and aggregation in response to collagen, is caused by defects in GPVI.
