General description

Platelet derived growth factor (PDGF) was discovered as a major mitogenic factor present in serum but absent from plasma. It was found to be secreted from the ฮฑgranules of platelets activated during the coagulation of blood to form serum. Subsequent studies have demonstrated that PDGF is not one molecule but three, each a dimeric combination of two distinct but structurally related peptide chains designated A and B. The dimeric isoforms PDGFAA, AB and BB are differentially expressed in various cell types and their effects are mediated through two distinct receptors, termed ฮฑ and ฮฒ. The PDGFs are stored in platelet ฮฑ-granules and are released upon platelet activation. The PDGFs are involved in a number of biological processes, including hyperplasia, chemotaxis, embryonic neuron development, and respiratory tubule epithelial cell development. Two distinct signaling receptors used by PDGFs have been identified and named PDGFR-ฮฑ and PDGFR-ฮฒ. PDGFR-ฮฑ is high-affinity receptor for each of the three PDGF forms. On the other hand, PDGFR-ฮฒ interacts with only PDGF-BB and PDGF-AB. Recombinant human PDGF-BB is a 24.3 kDa disulfide-linked homodimer of two B chains (218 total amino acids).
