General description
These indicators consist of Bacillus stearothermophilus (ATCC 7953) spores impregnated on paper strips, individually placed into envelopes. Bacillus stearothermophilus is a thermophilic species which can grow at 65C and above. The spores are highly resistant and are used to monitor autoclave performance. These indicators as specified by U.S. military specification MIL-S-36586 are GMP requirements of U.S. FDA.
Directions: Place indicators so that they are exposed representative to the steam as the media and the equipment. For the routine evaluation of each radiated lot a standard procedure should be established. After steam sterilization using rigid aseptic technique and open the envelope with the strip inside. Inoculate strip after autoclaving in a tube with sterile CASO Broth (22098) and incubate at 55-60C up to 7 days according USP. Today it is up to date in most laboratories to incubate for 48 hours according new stability studies. Simultaneously an unexposed spore strip is inoculated separately in another tube with sterile CASO Broth.
Quality control: Cultural response observed after 7 days at 55-60C in CASO Broth (22098): Spore Strip Results - Exposed to steam shows no growth; Unexposed shows luxuriant growth.
