Biochem/physiol Actions

La anfotericina B es un antibiótico antifúngico poliénico procedente del género Streptomyces, que se utiliza clínicamente para tratar las infecciones fúngicas sistémicas. Tiene una gran afinidad por los esteroles, especialmente los ergosteroles, de las membranas celulares tanto fúngicas como bacterianas. El uso normal en el mantenimiento de los cultivos celulares antifúngicos es de 2,5 mg/l con penicilina y estreptomicina en el medio.
La anfotericina B muestra su máxima eficacia contra hongos y levaduras. Forma canales transmembranarios al unirse a los esteroles asociados a las membranas, lo que induce la fuga de moléculas pequeñas. La anfotericina B provoca toxicidad a pesar de ser un antibiótico eficaz para tratar las micosis.

La anfotericina B permanece activa durante 3 días en cultivo a 37°C.

La anfotericina B es una lactona macrocíclica con una cadena polihidroxílica hidrófila y una cadena lipófila de hidrocarburo poliénico.

Este producto se suministra como 250 μg/ml de anfotericina B en agua desionizada. Es una disolución que contiene anfotericina B y los siguientes componentes dirigidos a mejorar su solubilidad: desoxicolato de sodio, cloruro de sodio y fosfato de sodio
Mantenga el recipiente herméticamente cerrado y en un lugar bien ventilado. Sensible a la luz.

Preparation Note

No es inusual que este material forme un precipitado en disoluciones acuosas. La mezcla de la disolución redisolverá la mayor parte del precipitado, pero si no se resolubiliza del todo, todavía podrá utilizarse la anfotericina B.
