Application

Follicle stimulating hormone (FSH) from human pituitary has been used for-
  • in vitro luteinization of Macaque granulosa cells
  • luteal cell proliferation assay
  • culturing of preantral follicle cells obtained from mice
  • in vitro maturation of immature cumulus-oocyte complexes (COCs) obtained from mice and
  • culture of RAW264.7 cell line

Biochem/physiol Actions

FSH (follicle stimulating hormone) in involved in the growth of ovarian follicles and steroidogenesis of ovaries. FSHR (follicle-stimulating hormone receptor) interacts with FSH in the ovaries and determines the responsiveness to this hormone. Depressed levels of FSHR result in poor responsiveness and influence folliculogenesis. In the Caucasian population, certain polymorphisms in the FSHR gene are more commonly found in patients with polycystic ovary syndrome (PCOS). In the male fetuses and neonates, it facilitates the proliferation of Sertoli cells. In pubescent males, it stimulates spermatogonia to undergo mitosis.FSH upon binding to its receptor FSH-R positively regulates G-protein coupled receptors/cyclic adenosine monophosphate (cAMP)/protein kinase A pathways to cascade intracellular signals.
The α-chain is not active; biological specificity is attributed to the β-chain. Induces maturation of Graafian follicles of the ovary; promotes development of germinal cells in males; activates cytosolic tyrosine kinase.

General description

The α chain of FSH has 89 amino acids and the β chain has 115 amino acids. The α chain is not active. FSH belongs to glycoprotein hormone family. It is secreted by the gonadotropic cells of pituitary gland.

  5. For Product F4021, Follicle Stimulating Hormone, do you have information on reconstitution, solution storage and potency?

    Product F4021-1VL is packaged as 50 units/vial. Each vial contains roughly 35-36 milligrams solid. Our solubility testing is done at 50 mg/mL, which yields a slightly hazy solution. It may be more convenient to use units/mL for your experiment as it will be more precise. If you decide to try to weigh out the material, please realize that small quantities can be difficult to weigh accurately.For Product F4021, FSH from human pituitary, we recommend solution storage for up to one month at 2-8°C. Unfortunately, we do not have any Sigma data regarding the stability of the porcine hormone.In the absence of information, we would recommend that solutions of Product F4021 not be frozen. If you do decide to store the product at -20°C we would suggest storing it in single use aliquots to avoid numerous freeze/thaw cycles.For the human FSH product we list an approximate activity of 7000 International Units/milligram. This should provide you with a starting point for a conversion calculation. For the porcine product we do not provide a conversion for units to mass. We provide 50 Armour units per vial. The unit definitions are also slightly different. International units are based on an international standard (IRP-68/140) while Armour units are based on Armour FSH standard G-94. According to the literature, 1 Armour unit can contain 9.4-14.2 international units.

  6. Do you have information on the solution stability of Follicle Stimulating Hormone (FSH) from pituitary?

    For information on the solution stability of FSH (and much more), refer to Chem. Soc. Rev., 2(4), 355-395 (1973).From page 369:  "The stabilities of the most highly purified preparations of certain hormonal glycoproteins have presented some problems, e.g. follicle-stimulating hormone preparations are often found to be stable in frozen solution but lose activity dramatically on concentration and freeze drying." Based on that information, we would recommend that if you need to freeze solutions of FSH, you should flash-freeze the solution (using either liquid nitrogen or a dry ice-alcohol bath), and store the solution in the ultra-cold (-70 or -80°C) freezer. Since FSH does have some solution stability concerns, simply placing the solution in a -20°C freezer without first flash-freezing may reduce the activity.
