Application

L'amphotéricine B est un antibiotique antifongique polyène extrait du bactérie du genre Streptomyces. Elle a une grande affinité pour les stérols, particulièrement pour les ergostérols, présents dans la membrane des cellules bactériennes et fongiques. L'usage normal en maintenance de cultures cellulaires antifongiques est de 2,5 mg/l avec utilisation de pénicilline et de streptomycine dans le milieu.

Biochem/physiol Actions

L′amphotéricine B se fixe aux stérols, formant ainsi des pores dans la membrane et provoquant la libération de petites molécules. Ce produit est particulièrement efficace contre les champignons et les levures.

Disclaimer

L'amphotéricine B demeure active pendant 3 jours en culture à 37 °C. À -20 °C, et protégé de l'air et de la lumière, le produit demeure actif pendant 5 ans.

Ce produit est une suspension colloïdale d'amphotéricine B, utilisant du désoxycholate de sodium comme agent solubilisant. La formulation est ~45 % d'amphotéricine B et de 35 % de désoxycholate de sodium dans du phosphate de sodium et du chlorure de sodium.
Conserver le récipient bien fermé et dans un endroit sec et bien ventilé. Le produit est sensible à la lumière et à l'humidité. Conserver dans un endroit sec.
Le poids solide réel du produit conditionné est calculé à partir de sa pureté UV.

Packaging

L'amphotéricine B solubilisée est conditionnée dans un flacon en verre, le contenu se trouvant dans un cône intérieur intégré.

Preparation Note

L'amphotéricine B est soluble à raison de 19,6-20,4 mg/ml dans l'eau, à pH naturel sans chauffage, donnant une solution transparente jaunâtre à légèrement trouble.
Les solutions aqueuses ne peuvent pas être stérilisées par filtration en raison de leur faible solubilité. Si elle est reconstituée dans de l'eau stérile, il n'est pas nécessaire de recourir à une filtration stérilisante.

Elle est également soluble dans le DMSO (30-40 mg/ml) et dans le diméthylformamide (2-4 mg/ml).

    We recommend to use Product No. A9528 at 5.6 mg solid per Liter.  Cell culture tested amphotericin B solubilized preparation is approximately 45% amphotericin B, 35% sodium deoxycholate, with the balance being sodium phosphate. The amphotericin B is at least 80% amphotericin B and 3-5% amphotericin A by HPLC.  The exact amount of amphotericin B in the product can be found on the lot specific Certificate of Analysis under Potency.
