Application

Excellent réactif pour maintenir les groupements SH à l′état réduit ; réduit de manière quantitative les disulfures. Lorsqu′il est ajouté aux tampons pour échantillons, le DTT permet de réduire les liaisons disulfure avec les protéines avant une analyse SDS-PAGE. Le DTT peut également être utilisé pour réduire le pont disulfure de la N,N′-bis(acryloyl)cystamine, un agent réticulant, afin de démanteler la matrice d′un gel de polyacrylamide. Le DTT est moins caustique et moins toxique que le 2-mercaptoéthanol. Généralement, la concentration de DTT nécessaire (100 mM) est sept fois moins élevée que celle du 2-mercaptoéthanol (5 % v/v, 700 mM).
