Présente également des effets immunosuppresseurs, par exemple avec la polyarthrite rhumatoïde.

Également efficace dans le traitement des parasites de la malaria Plasmodium vivax résistant à la pyriméthamine.
Le méthotrexate hydraté a été utilisé :
  • Comme inhibiteur du dihydrofolate (DHFR) dans une culture destinée à un essai de formation de sphère.
  • Comme constituant d′un milieu de Eagle modifié par Dulbecco (DMEM) pour la sélection de cellules transfectées.
  • Dans des essais de viabilité et de co-culture.

Le méthotrexate est un antagoniste de l′acide folique.
Le méthotrexate sert à inhiber la dihydrofolate réductase (DHFR) et à sélectionner la surexpression de DHFR.
