Application

Ces comprimés ont été optimisés et testés pour un usage général. Ils ont été utilisés dans diverses applications, notamment pour recueillir des trachées conjointement avec un tampon de radio-immunoprécipitation glacé afin d' identifier le devenir des cellules basales trachéales, et pour homogénéiser des tissus de poumon de rat destinés à une analyse par western blotting.
Les comprimés d'inhibiteurs de protéases SIGMAFAST ont été utilisés :
  • pour laver du sperme brut en vue de l'analyse de protéines membranaires.
  • dans une solution destinée à incuber des lymphocytes T CD4+ pour obtenir des lysats cellulaires totaux devant être analysés par western blotting.
  • comme constituant d'un tampon à base de NP-40 pour homogénéiser le tissu de moelle épinière centré sur l'épicentre d'une lésion et réaliser une analyse par western blotting.
  • comme constituant d'un tampon pour un essai de radioimmunoprécipitation (RIPA) afin d'homogénéiser le tissu de moelle épinière centré sur le centre d'une lésion et détecter de la neuréguline-1 bêta 1 (Nrg-1ß1) dans des échantillons de moelle épinière avec un test ELISA (essai immuno-enzymatique).

General description

Il n'est pas conseillé d'utiliser les comprimés d'inhibiteurs de protéases SIGMAFAST avec les produits HIS-Select® ou d'autres produits de chromatographie d'affinité sur ions métalliques immobilisés (IMAC). Ces comprimés contiennent de l'EDTA, un chélateur de métaux pouvant potentiellement extraire les ions métalliques contenus dans ces produits fonctionnant par affinité.
Les comprimés d'inhibiteurs de protéases SIGMAFAST™ constituent une formulation hydrosoluble contenant un cocktail d'inhibiteurs de protéases à large spectre, incluant de l'AEBSF, de l'E-64, de la bestatine, de la leupeptine, de l'aprotinine et de l'EDTA. Un comprimé permet de préparer 100 ml de cocktail et peut inhiber les protéases présentes dans un extrait cellulaire de 20 g maximum.

Preparation Note

Un comprimé donne 100 ml de cocktail. Il est recommandé d'utiliser 1 ml pour l'inhibition de protéases équivalant à 1 mg de pancréatine USP. Il est recommandé d'utiliser 1 comprimé pour l'inhibition des protéases présentes dans un extrait cellulaire de 20 g maximum.

    S8820 is the tablet version of P2714.  

  6. Can Product S8820, SIGMAFAST™ Protease Inhibitor Tablets, be used with with HIS-Select™ products or any other immobilized metal affinity chromatography (IMAC) products?

    The tablets contain EDTA, a metal chelator, which may remove metal ions from these affinity products. The suggested protease inhibitor for the HIS-Select products is either Product No. S8830 (water soluble tablet) or P8849 (DMSO solution).

  7. Can Product S8820, SIGMAFAST™ Protease Inhibitor Tablets, be used with ELISA?

    Yes, because it is water based.

  8. How should I dissolve Product S8820, SIGMAFAST™ Protease Inhibitor Tablets?

    You can dissolve the product in 10 ml of water to make a 10X stock solution and then dilute 1:10 for use.

  9. What is the concentration of the inhibitors in Product S8820, SIGMAFAST™ Protease Inhibitor Tablets?
