Biochem/physiol Actions

Cet inhibiteur agit contre la trypsine ; il présente une activité réduite envers la chymotrypsine et la plasmine. Il inhibe également les protéases dont le mécanisme est similaire à celui de la trypsine, la kallikréine plasmatique et le facteur de coagulation X. L′inhibiteur de trypsine n′agit pas contre les métalloprotéases, les kallikréines tissulaires, les protéases acides ou les thio-protéases. Cet inhibiteur agit en formant un complexe d′une façon stœchiométrique 1 à 1 avec le site actif de la protéase, suivi du clivage d′une seule liaison arginine-isoleucine présente dans l′inhibiteur. L′inhibition est réversible et dépendante du pH.

Preparation Note

L′inhibiteur de trypsine est soluble dans l′eau et les tampons phosphate à 10 mg/ml. Il est soluble dans des solutions salines équilibrées à 1 mg/ml et dans les milieux sans sérum. Les solutions d′une concentration supérieure à 10 mg/ml peuvent être troubles et de couleur jaune à ambrée. Après le traitement des cellules à la trypsine, les resuspendre dans une solution d′inhibiteur de trypsine à 1 mg/ml pour chaque ml de solution de trypsine utilisée pour la dissociation. La suspension cellulaire doit alors être centrifugée à 1000 tr/min pour former un culot de cellules.

Les solutions maintiennent leur activité lorsqu′elles sont conservées à court terme entre 2 et 8 °C. Les solutions sont stables en aliquotes congelés à -20 °C.

Analysis Note

One mg of trypsin inhibitor will inhibit a minimum of 1.0 mg Trypsin of activity ~10,000 BAEE units/mg trypsin.

    Soybean trypsin inhibitor is a mixture of the three types. We have not characterized our trypsin inhibitor products in terms of how much of each of the three types may be present. All three have a molecular weight of between 20,000 and 20,200 daltons, so one might use 20,100 daltons as an average.The three types vary in the amino acids at a few positions. The detailed summary of the three types (or variants) was published in the following linked reference:     J. Biochem. (Japan), 98, 435-448 (1985).Note that the authors use Tia, Tib and Tic to indicate the three variants.
