General description

Insulin-like growth factor 2 (UniProt: P01346; also known Insulin-like growth factor II, IGF-II, Multiplication-stimulating activity, MSA, Multiplication-stimulating polypeptide) is encoded by the Igf2 (also known Igf-2) gene (Gene ID: 24483) in rat. This gene encodes a member of the insulin family of polypeptide growth factors that is primarily involved in fetal development, influencing cellular proliferation and differentiation. It is an imprinted gene, with expression predominantly from the paternally inherited allele. It is regulated by multiple epigenetic mechanisms, including differential methylation and transcription factor binding at the IGF2-H19 locus on chromosome 11p15. It is a candidate gene for eating disorders. IGF-II is a widely expressed, secreted protein that is synthesized with a signal peptide (aa 1-24), propeptide (aa 92-180) which is subsequently cleaved off to produce the mature protein (aa 25-91). Human IGF-II shares similar structural features with IGFI, including a 62% sequence homology. Alternatively spliced transcript variants, encoding either the same or different isoform, have been found for this gene. In human plasma, IGFI and IGFII are associated with IGF-binding proteins that transport the polypeptides and partially regulate their actions in vivo. In addition to the insulin receptor, IGFII binds to two forms of IGF receptors, both of which are widely distributed in different tissues and cultured cells. IGFII is mitogenic for a variety of cultured cells, including mouse 3T3 cells,12 normal rat kidney cells, human or chicken fibroblasts and MCF-7 human breast carcinoma cells. Dysregulation of IGF-II expression has been implicated in various diseases, including overgrowth syndromes such as Beckwith-Wiedemann syndrome and Silver-Russell syndrome, as well as in several cancers, where IGF-II promotes tumor growth and survival. Specificity

Clone S1F2 is a mouse monoclonal antibody that specifically detects IGF-II. This antibody has less than 10% cross-reactivity to human IGF-I.
