Preparation Note
For maximum recovery of product, centrifuge the vial prior to removing the cap.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variabillity in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
Analysis Note
Acid extracted HL-60 cells treated with VP16
Western Blot Analysis:
1:1,000-1:10,000 dilutions of this lot detected phospho-Histone H2B in acid extracted HL-60 cells treated with 20μg/mL VP16 for 18 hours. A 1:250-1:2,000 dilution of a previous lot also detected phospho-Histone H2B acid extracted proteins from colcemid-arrested HeLa cells.
Note:
The optimal antibody dilution of this antibody must be empirically determined. Use of this antibody at dilutions which are too low, will result in non-specific detection of Histone H2B and other unknown proteins in western blots of acid extracts from untreated cells.
