General description
A cell-permeable hydroxyanthraquinone compound that binds directly to the phosphoryl Tyr705 binding site of the STAT3 monomer and acts as selective and reversible inhibitor of STAT3 signaling. Shown to block STAT3 phosphorylation and its subsequent nuclear localization and DNA binding, but has no effect on IL-4, interferon-α and interferon-γ-induced STAT1, 2, 4, 6 and pJAK2, PI3-Kinase, or ERK signaling. Does not affect the activity of over 21 other closely related protein kinases (IC50 >50 µM). Also shown to induce apoptosis in U266 and ARH-77 multiple myeloma cell lines (IC50 = 490 nM and 1.96 µM, respectively), and suppress hepatocellular carcinoma tumor growth in xenograft mouse model (5 mg/kg, i.v).
