General Cell Biology

General cell biology is rapidly expanding its use of flow cytometry as a tool to examine pathways and propose biologically-relevant mechanisms of action in pathologies such as cancer. As shown below, many new applications are currently available to aid research in this area.

Flow cytometry analysis of phospho-EGFR (Y1173) in A431 cells treated with gefitinib. After treatment with gefitinib in a dose dependent manner, an IC50 value derived from MFI values was determined to be 31nM (A). Using InCyte™ software, the levels of EGFR inhibition were visualized and quantitated in a heatmap (B).

Cell Signaling

Signal transduction pathways lead to diverse outcomes, such as apoptosis, cell differentiation, cell growth and cell proliferation, all of which have been extensively studied in the process of developing therapies for various cancers and autoimmune disease. Cross-talk among signaling pathways adds an extra dimension of complexity when analyzing physiological consequences of a pathway of interest. However, there are some key nodes at which multiple signals are integrated. Multiparametric flow cytometry provides researchers the power to monitor these key intracellular ‘checkpoints’ simultaneously, enabling analysis of complicated cell events.

Detection of live and dead GFP transfected and untransfected cells. A mixture of untransfected cells (CHO), GFP transfected cells (C166) and C166 cells treated with Celastrol (to generate dead transfectants) was incubated with 5μ/mL Pl solution and then acquired on the Guava PCA-96 AFP System.

New customized instrument formats allow green and red fluorescent protein detection on the easyCyte™ system. Amnis® systems also have the ability to detect a variety of fluorescent proteins.
