通常はFasを発現するヒト細胞で細胞溶解活性を示すことにより評価されます。ヒトFasをコードしているcDNAでトランスフェクトされたマウスWR19L細胞およびL929細胞は、この抗体に反応してアポトーシスを起こします。

アポトーシスアッセイ分析：
15～20 µg/mLのこの抗体は、処理後24時間の時点でヒトJurkat細胞のアポトーシスを83%の死亡率で最大限に誘導できます。

Application

この抗Fas抗体（ヒト、活性）、クローンCH11を用いたFasの検出は、フローサイトメトリー（FC）、免疫細胞染色（IC）およびウェスタンブロッティング（WB）への使用が公表およびバリデーションされています。
ウェスタンブロッティング：
0.5～2 μg/mLを使用、Raji細胞ライセートでFasを検出しました。
免疫細胞染色：
5～10 μg/mLを使用、4%ホルマリン／2% 酢酸で固定したHeLa細胞でFasを検出しました。
フローサイトメトリー：
独立した研究所において20 μg/mLの抗Fas、クローンCH11を使用してテストしています（Yonehara, S., 1989; Kobayashi, N., 1990）。

この抗体はTNFを認識せず、マウスFasと交差反応しません。 Fasリガンドは、ヒト、マウスおよびラット系でアポトーシスを誘導します。
この抗体は、骨髄性細胞、Tリンパ芽球様細胞、および2倍体線維芽細胞を含むさまざまなヒト細胞で発現する、ヒト細胞表面抗原Fas、Mr 43 kDaを認識します。

General description

43 kDa
Fas/APO-1/CD95（36 kDa）は、膜貫通受容体のファミリーである腫瘍壊死因子（TNF）受容体スーパーファミリーのメンバーです。Fasは、アポトーシス細胞死の重要なメディエーターであることと、炎症に関与することが分かっています。Fasリガンド（Fas-L）の結合は、標的細胞膜内のFasの三量化を誘導します。Fasの活性化は、Fasのデスドメインと、デスドメインを持つFas関連タンパク質（FADD）との間の相互作用を介してFADDの動員を引き起こします。プロカスパーゼ8は、FADDのデスエフェクタードメイン（DED）とプロカスパーゼ8との間の相互作用を介してFas-結合FADDと結合し、カスパーゼ8の活性化をもたらします。活性化したカスパーゼ8は、他のプロカスパーゼを切断（活性化）し、最終的にアポトーシスにつながるカスパーゼカスケードを実質的に開始します。カスパーゼは核ラミンを切断し、核を崩壊させてその正常構造を失わせます。Fas-誘導アポトーシスは、FLICE阻害タンパク質（FLIP）、Bcl-2、またはサイトカイン反応修飾物質A（CrmA）のいずれかにより、いくつかのステージで効果的に阻止することができます。

生物活性
この抗体は、Fasを発現するヒト細胞で細胞溶解活性を示します。ヒトFasをコードしているcDNAでトランスフェクトされたマウスWR19L細胞およびL929細胞は、この抗体に反応してアポトーシスを起こします。
