General description

A cell-permeable, practically non-fluorescent (Φ0 = 0.001) coumarin cis-acrylate derivative that exhibits a 470-fold increase in fluorescence property (Φ = 0.47; Ex. 400 nm, Em. 465 nm) upon an 1:1 Michael adduct formation with thiol/sulfhydryl and acts as a fast-reacting (t1/2 = 3 msec with 50 µM probe and 50 mM 2-ME at t = 0), highly sensitive (Detection limit ≤0.5 nM GSH with 100 nM probe), and thiol-specific fluorogenic probe, displaying high reactivity toward 2-ME, GSH, and Cysteine, but not non-thiol-containing GSSG or Cystine. A more sensitive alternative to Coomassie Blue in staining of BSA on SDS-PAGE gels and has been applied successfully in fluorescent labeling cellular thiols in intact HEK293 cells as well as in detecting glutathione reductase activity in cell-free enzymatic assays. ThioGlo1, ThioGlo3, and ThioGlo5, in comparison, exhibit much lower fluorescence quantum yield increases upon reaction with thiol (61-, 156-, and 28-fold increase, respectively).

Other Notes

Packaged under inert gas. Ok to freeze. Protect from light
Use only fresh DMSO that for reconstitution. Following reconstitution, aliquot and freeze (-20°C). Stock solutions are stable for up to 3 months at -20°C.
