General description

アセチルコリン（ACh）は、運動ニューロン、自律神経節前ニューロン、副交感神経節後ニューロン、さまざまな脳領域、一部の新興ニューロン様幹細胞に共通する神経伝達物質です。Achの代謝は比較的単純で、合成にはコリンアセチルトランスフェラーゼ（ChAT）、分解にはアセチルコリンエステラーゼ（AChE）という2つの酵素のみが関与しています。また、アセチルコリンは神経伝達以外のニューロンではほとんど機能しておらず、ニューロン特異的であると考えられます。コリン作動性ニューロンのみがかなりの量のChATを有するため、抗コリンアセチルトランスフェラーゼは有用な特異的マーカーになると考えられます。

Application

免疫沈降：
1：200希釈で使用、脳および胎盤から68 kDaの活性ChATを免疫沈降しました。また、ヒトやおそらく他の哺乳類の中枢神経系の一部のコリン作働性神経において、ChATと共発現している（がChAT活性はない）27 kDaのタンパク質も沈殿させます。

最適な希釈濃度とプロトコルは、ご自身で決定してください。
抗コリンアセチルトランスフェラーゼ（ChAT）抗体は、コリンアセチルトランスフェラーゼ（ChAT）に対する抗体であり、IH、IH（P）、IP、WBでの使用が検証されており、製品の引用は70件以上になっています。

Analysis Note

コントロール
脳皮質組織。
正常脳皮質組織の免疫組織染色（パラフィン）で定常的に評価済み。

免疫組織染色（パラフィン）：
正常脳皮質のChAT（カタログ番号：AB143）染色。組織を処理しましたが、抗原は回収されませんでした。本抗体をIHC-Selectを用いて1：100に希釈しましたHRP-DABによる検出。 免疫反応性は、ニューロンの細胞体、樹状突起、軸索における染色パターン（褐色）として認められます。
