General description
MEF支持細胞のプレーティング
手順：
1.PMEF支持細胞を融解する前に、プレート／フラスコをゼラチン溶液でコーティングします。
2.PMEFバイアルを37°Cの水槽に入れて素早く融解し、15 mLチューブ（10 mLの温かいPMEF支持細胞培地がすでに含まれている）に移します。チューブを穏やかに転倒させて混ぜ、300 x gで4–5分間遠心します。
3.上清を除去してから、温かいPMEF支持細胞培地中で細胞ペレットを再懸濁します。
4.プレート／フラスコからゼラチン溶液を除去し、マウスESプロトコルガイドの表4.1で推奨されている濃度でPMEF支持細胞懸濁液を分注します。
5.PMEF支持細胞を37°Cで5% CO2により培養します。マウスESプロトコルガイドの図4A、BおよびCを、正しいPMEFの濃度と外観を推定するためのガイドとして
使用してください。ゼラチン化したプレートは12–14日間使用できます。
