Application

MystiCq® microRNA cDNA Synthesis Mixは、総RNAから開始される定量的ポリメラーゼ連鎖反応（qPCR）用のmiRNAの逆転写によるファーストストランドcDNA合成のために使用されています。

General description

MystiCq® microRNA cDNA Synthesis Mixキットには、総RNAまたは濃縮サンプルまたはマイクロRNAから開始して、成熟マイクロRNAをqPCR用のcDNAテンプレートに変換するために必要な全ての成分が含まれています。 マイクロRNAは自然にはポリアデニル化されないため、cDNA合成の最初の段階は、ポリ(A)ポリメラーゼ反応によりマイクロRNAをポリアデニル化することです。 次に、ReadyScript逆転写酵素、オリゴ-dTアダプタープライマー、およびその他の必要な試薬を添加して、ポリアデニル化したマイクロRNAをcDNAに変換します。 アダプタープライマーには、5′末端に、MystiCq microRNA qPCR Assay Systemのもうひとつの要素であるMystiCq Universal PCR Primerの配列と相補的なユニークな配列が含まれています。キットには、ほとんどのヒト組織で広く発現している低分子核小体RNAであるSNORD44に特異的な、Positive Control Primerも含まれています。 キットには、十分な量のポリ（A）Tailing Bufferと、MicroRNA cDNA Reaction混合物が含まれているため、ポリ（A）ポリメラーゼや逆転写酵素調節反応を用いる必要はありません。個々のマイクロRNAを分析するには、MystiCq Universal PCR Primer（アダプターのプライマー配列を標的にします）を、MystiCq®microRNA qPCR Assay PrimerおよびMystiCq microRNA SYBR®Green qPCR ReadyMixと併用します™。
