酸性線維芽細胞増殖因子(FGF-1)と塩基性線維芽細胞増殖因子(FGF-2)は多くの組織にみられる遍在性のサイトカインです。内皮細胞など中胚葉や神経外胚葉由来の複数の細胞型に影響を及ぼします。FGFタンパク質は、155～268位のアミノ酸残基からなる小型のペプチドです。異なるファミリーメンバー間の配列の同一性の程度は、約120個のアミノ酸からなる「中央ドメイン」で30～60%です。このドメインは、共通の三次構造とヘパリンへの結合能を、FGFに付与します。分泌されるFGFは、細胞表面チロシンキナーゼFGF受容体(FGFR;6、7)に結合し活性化することによって、標的細胞にシグナルを伝達します。いくつかの動物モデルで、胚発達中および成体の生理におけるFGFとFGFRの機能が、機能獲得および機能喪失実験によって検討されました。これらの研究により、FGFが発達イベントの主な調節因子として機能することが示されました。
