General description
Phosphatidic acid (PA), a minor constituent of biological membranes is a simplest glycerophospholipid. It has glycerol backbone with phosphate present at C-3 position. The C-1 and C-2 positions are esterified with fatty acids.
Application
10:0 PA may be used:
- as a component in the lipid bilayer to study how lipid geometry acts in coat protein I (COPI) vesicle fission
- to study its effects on intracellular Ca2+ concentration ([Ca2+]i) in C6 rat glioma and L2071 mouse fibroblast cells
- to activate protein kinase C (PKC)
- as a cell-free activator in membrane and cytosolic fractions for nicotinamide adenine dinucleotide phosphate (NADPH) oxidase assay
Biochem/physiol Actions
Phosphatidic acid (PA) serves as a lipid second messenger. It can control membrane shape. It is known to participate in cell proliferation, vesicular trafficking, cytoskeletal organization and signal transduction. PA can stimulate negative curvature in lamellar lipid structures. It can block the enzymatic activity of protein phosphatase 1 (PP1).
