General description

Estrogen Receptor alpha is a 65 kDa protein and a member of the steroid family of nuclear receptors. Estrogen Receptor alpha is a ligand-activated transcription factor that, when bound to estrogen, induces a conformational change that allows dimerization and binding to estrogen response element sequences. When bound to DNA, Estrogen Receptor alpha can positively or negatively regulate gene transcription. Like other steroid hormone receptors, Estrogen Receptors are intracellular proteins. Estrogen Receptors play an important role in regulating mammary gland growth and differentiation. Breast tumors that are ER positive may also show a more favorable response to anti-estrogen therapies.
~58 kDa observed. UniProtKB/Swiss-Prot P06211 (ESR1_RAT) lists a molecular weight of 67 kDa A molecular weight between 52-58 kDA due to variant isoforms may be observed in some cell lysates (Al-Bader, M.D. (2006). Reprod Biol Endocrinol. 4:13.; Geisler, J.G. et al. (2002). Diabetes. 51(7):2158-2169.). Uncharacterized bands may be seen at a molecular weight of ~10 kDa, ~35 kDa, ~41 kDa, ~105 kDa, and ~220 kDa in some cell lysates.

Immunogen

Epitope: C-terminus.
KLH-conjugated linear peptide corresponding to the C-terminus of rat Estrogen Receptor alpha.

Application

  • Tested applications
  • Immunohistochemistry (Paraffin) Analysis: A 1:400 dilution from a representative lot detected Estrogen Receptor in human stomach, human uterine cancer, human breast cancer and human cervical cancer tissue sections.
  • Immunoprecipitation Analysis: 4 µg from a representative lot immunoprecipitated Estrogen Receptor in MCF-7 cell lysate.

A major focus of breast cancer research is to understand the mechanisms responsible for disease progression and drug resistance. Toward that end, it has been found that approximately two thirds of all human breast carcinomas overexpress the Estrogen Receptor α (ERα) protein and it remains the primary pharmacological target for endocrine therapy1,2. The normal cellular function of ERα is as a transcription factor that mediates a wide variety of physiological processes, many of which are dependent upon phosphorylation of the receptor at specific amino acid residues3,4. Indeed, ERα is known to be phosphorylated at a multitude of different sites, yet how these all correlate to disease remains unclear5. Here, we interrogated multiple sites of ERα for phosphorylation status by screening an extensive panel of different breast cancer patient samples and other non-breast cancer tissue microarray (TMA) slide samples to determine their relevance to disease.
