General description
Autophagy is a catabolic process by which cell delivers cytoplasmic constituent into lysosome for protein degradation. 16 ATG genes are essential for autophagosome preparation out of which Atg7 is the one required to activate ubiquitin-like proteins Atg12 and Atg8. Atg7 is also crucial for amino acid supply in neonates, as well as for bulk degradation of proteins and organelles in mice during starvation. Anti-ATG7 antibody can be used in immunohistochemistry and immunoprecipitation. Rabbit anti-ATG7 antibody reacts specifically with Atg7 of human, mouse and rat.
Immunogen
synthetic peptide corresponding to amino acids 38-50 of human ATG7, conjugated to KLH via a C-terminal cysteine residue. The corresponding sequence is identical in mouse and rat.
Application
Anti-ATG7 antibody can be used in western blotting and immunoblotting.
Anti-ATG7 antibody produced in rabbit has been used:
- for quantitative real-time polymerase chain reaction of autophagy-related genes
- in immunofluorescence and immunoblotting
- in in vivo acetylation/deacetylation of autophagy proteins
Rabbit anti-ATG7 antibody has been used for immunoblotting assays. The antibody is also suitable for use in immunoprecipitation applications at 1-2μL using human U87 cell lysates.
Rabbit anti-ATG7 antibody was used for immunohistochemistry on sections of mouse midbrain fixed in 4% paraformaldehyde at 1:2000.
Biochem/physiol Actions
Autophagy related 7 (ATG7) regulates caspase mediated autophagy and apoptosis. This has a significant effect on tumor progression and ATG7 acts as a tumor suppressor. ATG7 is known to be associated with many cancer types, hence it is considered as a target for cancer therapy. Loss of ATG7 gene contributes to liver carcinogenesis. The ATG7 gene encodes E1-like enzyme essential for lipidation and conjugation systems : ubiquitin-like microtubule-associated protein 1A/1B-light chain 3 (LC3) and autophagy related 12 (ATG12) respectively.
