General description
Histone H1 is part of high-order chromatin structure and is a linker protein associated with nucleosome core particle. Histone H1 is mapped to human chromosome 6. Histone H1 exists in different isoforms. Histone H1 comprises of N-terminal hydrophobic rich region, central globular domain and a C-terminal with intrinsically disordered regions.
Biochem/physiol Actions
The C-terminal regions of histone H1 is essential for binding to chromatin. The placement of H1 is necessary for nucleosome positioning. Histone H1 maintains chromosome architecture and stabilises compaction of nucleosomes during interphase. H1-null mutants display increased rate of chromatin aberration and growth defects. Histone H1 modulates embryogenesis and DNA repair. The H1 histone undergoes posttranslational modification including phosphorylation, methylation and acetylation, each of which carry out different physiological functions. Histone H1 and its variants contribute to the pathogenesis in cancer, Alzheimer′s disease, male infertility and immunity related disorders.
