      DescriptionPurified rabbit polyclonal antibody. Recognizes Fas ligand.
      BackgroundThe ligand for fas/APO-1/CD95 was identified in a cytotoxic T cell hybridoma that could lyse cells expressing fas/APO-1 but not cells lacking this receptor. Subsequent work directly demonstrated that triggering through the fas molecule by the fas ligand was responsible for the induction of apoptosis. The fas ligand is a member of the TNF family of proteins and is classified as a type II membrane protein. The highest degree of homology with TNF occurs in the extracellular domain which corresponds to the COOH terminal portion of the fas protein. The fas ligand has four potential N-glycosylation sites which appear to be variably used. Because of the variation in glycosylation, the full-length protein migrates as several distinct molecular weight species by SDS-PAGE. In addition to the full-length protein of ~35-43 kDa, it has been shown that the fas ligand is also cleaved from the surface of the cell and released as a soluble form approximately 26 kDa in size. There is still debate as to whether the protein is actively cleaved or is simply released/shed from the cell membrane. The chromosomal location of fas ligand has been mapped to 1q23 and the gene is split into 4 exons spanning a distance of about 8.0 kb. Involvement of fas/fas ligand has now been demonstrated as being a general mechanism for the action of cytotoxic T lymphocytes.
