Application

L′IPTG est couramment utilisé dans les procédures de clonage nécessitant une induction de l′activité β-galactosidase. Il est utilisé en conjonction avec X-Gal ou Bluo-Gal dans la sélection par couleur bleue ou blanche des colonies bactériennes recombinantes dans lesquelles l′expression de l′opéron lac d′Escherichia coli est induite. L′IPTG fonctionne en se liant au répresseur lacI et en modifiant sa conformation, ce qui empêche la répression du gène lacZ qui encode la β-galactosidase.

Preparation Note

Une solution mère (0,1 M) est préparée en dissolvant de l′IPTG dans de l′eau, suivi par une stérilisation de la solution par filtration. La concentration finale d′IPTG dans les boîtes indicatrices est de 0,2 mM.
