胚性幹（ES）細胞は、着床前哺乳類胚の内細胞塊（ICM）に由来する全能細胞であり、in vitroでの無制限の未分化増殖が可能です[1,2,3]。 未分化マウスES細胞は、サイトカイン、白血病阻止因子（LIF）またはMillipore独自のES細胞培養試薬ESGROを含む培地において、in vitroでの長期維持が可能です[4,5]。 未分化状態のES細胞は、アルカリホスファターゼ（AP）の高レベルの発現[6]、SSEAおよびTRA抗原を含む表面マーカーの発現、および転写因子Oct-4の発現によって特徴付けられます。

Milliporeのアルカリホスファターゼ検出キット（カタログ番号：SCR004）は、AP活性の測定によってES細胞分化の表現型を評価するための特異度と感度に優れたツールです。

Milliporeから個別に入手できるモノクローナル抗体はTRA-2-49（カタログ番号：MAB4349）とTRA-2-54（カタログ番号：MAB4354）であり、ES細胞によって発現されるAPの肝臓／骨／腎臓アイソザイムの検出を可能にします。 フローサイトメトリーでこれらの試薬を使用すると、ES細胞によるAP発現を定量評価できます[7]。

研究専用です。診断にはご使用になれません。

Preparation Note

アルカリホスファターゼ検出キットは、AP活性の測定に用いられる2つの構成要素から成ります。

2～8º Cで保管した場合、キット構成要素は使用期限まで安定です。 凍結したり高温に曝露したりしないでください。使用期限後、残っている試薬はすべて廃棄してください。
