General description

正常細胞においてJC-1色素はミトコンドリア基質中に濃縮し、そこで赤の蛍光を発する凝集体を形成します。アポト-シスなどのミトコンドリア膜電位を消散させる事象が起こると、JC-1のミトコンドリア内での蓄積が妨げられ、色素が細胞全体に分散し、赤色蛍光 (JC-1凝集物) から緑色蛍光 (JC-1モノマ-) へと変化します。本キットで染色した細胞の蛍光は、蛍光顕微鏡、蛍光光度計やフロ-サイトメトリ-で検出できます。

Application

ミトコンドリアの電気化学的ポテンシャル勾配 (Δψ) の消失は、アポト-シス初期に起こります。本キットは、親油性のカチオン色素であるJC-1を利用して、生細胞におけるミトコンドリア内膜電気化学的ポテンシャルの変化を検出するための、迅速で便利な方法です。
