置換バルビツル酸であるフェノバルビタールは抗てんかん薬であり、循環血中から容易には排除されません。γアミノ酪酸（GABA）によるニューロンシナプス後GABAA受容体の活性化に有利に働くことにより、シナプスニューロン膜を過分極します。また、フェノバルビタールは、新生児発作やてんかん重積状態の治療にも有効です。
鎮静薬、睡眠薬、抗痙攣薬で、GABA作動性活性を増強します。
