Application

パクリタキセルは、NIH3T3細胞抽出物の免疫共沈降法によるPREP2とチューブリンの相互作用を研究するために用いられています。ブタの脳から得られたチューブリンを安定化するためにも用いられています。

Biochem/physiol Actions

パクリタキセルは、強力な抗悪性腫瘍および抗有糸分裂性のタキサン系薬物であり、β-チューブリンのN末端と結合し、微小管を安定化させてG2/M期の細胞周期を停止させます。

Preparation Note

パクリタキセルはDMSOに50 mg/mLで溶解すると数ヶ月間冷凍保存できます。メタノール (エステル交換を受ける)、アセトニトリルおよびエタノールにも可溶です。パクリタキセルは、弱アルカリ性のメタノール溶液中および強酸性のメタノール溶液 (メタノール:濃HCl＝1:1) 中で急速に分解します。50% Cremophor ELと50%無水エタノールの混合液にも溶解します。
