Biochem/physiol Actions

トリプシンは、リジンとアルギニン残基の C 末端側のペプチドを切断します。 この反応の加水分解速度は、切断箇所のどちらかの側に酸性残基がある場合に遅くなり、また、C 末端側にプロリン残基がある場合、加水分解は停止します。 トリプシン活性の至適 pH は 7 ～ 9 です。 トリプシンはまた、合成アミノ酸誘導体のエステルおよびアミド結合も切断できます。 EDTA は、トリプシンの作用するペプチド結合を曖昧にするカルシウムやマグネシウムイオンを中和するキレート剤として、トリプシン溶液に添加されます。 これらのイオンの除去は、酵素活性を上昇させます。

DFP、TLCK、APMSF、AEBSEF およびアプロチニンなどを含むセリンプロテアーゼ阻害剤は、トリプシンを阻害します。

トリプシンは、223 アミノ酸残基からなる単鎖ポリペプチドであり、トリプシノーゲンからの、Lys - lle 結合の位置で切断された N 末端側のヘキサペプチドの除去によって生成されます。 アミノ酸配列は、6 個のジスルフィド結合によって架橋されています。 これがトリプシンのネイティブフォームである、βトリプシンです。 βトリプシンは自己消化によって Lys - Ser 残基の位置で切断され、αトリプシンを生成します。 トリプシンは、セリンプロテアーゼファミリーに属します。
