トリプシンインヒビターは、エステラーゼ型酵素の単離のためのアフィニティーリガンドとして用いられています。
Phaseolus limensis（ライ豆）由来のトリプシン阻害剤は以下の用途に使用されています：
- ヒト血漿中のトリプシン阻害
- ウサギの骨格筋のタンパク質消化停止
- Lygus lineolaris（サビイロカスミカメ）から抽出された唾液腺酵素の阻害
- Agave tequilana（アガベ・テキーラナ）抽出酵素のタンパク質分解活性の阻害
Analysis Note
1 mgは、タンパク質1 mgあたり約10,000 BAEE単位の活性を有する≥0.8 mgのトリスピンを阻害します。
Biochem/physiol Actions
リマ豆由来のトリプシン阻害剤は、プロテアーゼ阻害剤のBowman-Birkファミリーに属しています。分子量は9 kDaで、9つのジスルフィド架橋を含み、非常に高い熱安定性を有しています。トリプシンとキモトリプシン結合サブドメインがあります。
General description
単量体は、みかけの分子量が約9,000 Daであり、濃度およびpH依存的に二量体化します。
Other Notes
1トリプシンユニットは、BAEEを基質として用いる場合、pH 7.6、25°C、1分間に、ΔA253が0.001となる酵素量です(反応液量 = 3.2 mL、光路長 = 1 cm)。
