      BackgroundThe MAD-related (MADR) family of proteins are intracellular signaling components for the TGFβ superfamily of ligands. Smad1 is the mammalian homologue of the Drosophila protein MAD. Smad1 mediates BMP signals in the cell. It is phosphorylated on serine residues at the C-terminus and forms heterodimers with another MAD family member, DPC4, in response to BMP2 and BMP4. In addition, MADR proteins that are distributed throughout the cell become localized to the nucleus following its activation.
