      Full-length, recombinant, human insulin receptor substrate p53 (IRSp53) fused to HisTag® and STag™ sequences at the N-terminus and expressed in E. coli. This preparation is qualified for use as a substrate for protein tyrosine kinases in in vitro assays. IRSp53 (a.k.a. IRS-58, IRSp53/58, BAIP2, BAIAP2, FLAF3) is a secretin receptor family protein induced by p53. Yeh, et al. cloned a hamster cDNA encoding a 53 kD BAI-associated protein that was tyrosine phosphorylated by the insulin receptor when expressed in mammalian cells. They termed the protein IRSp53 for "insulin receptor substrate p53". The protein contains 521 amino acids with a calculated molecular mass of 57 kDa and is highly expressed in the brain. IRSp53 is an adapter protein that links membrane-bound small G-proteins to cytoplasmic effector proteins and is necessary for CDC42-mediated reorganization of the actin cytoskeleton and for RAC1-mediated membrane ruffling. The protein appears to be localized primarily in the particulate fraction of cell lysates. It is phosphorylated by insulin receptor (InsR) in response to insulin treatment. Phosphorylated sites include Tyr337/Tyr505 by InsR and Tyr471 by Src and EGFR. In addition, the SH3 domain (aa 374-437) interacts with ATN1, BAI1, WASF1, WASF2, SHANK1, DIAPH1, and ENAH.
