Immunogen

Recombinant Human Brain Derived Neurotrophic Factor (BDNF).

Application

Anti-Brain Derived Neurotrophic Factor Antibody detects level of Brain Derived Neurotrophic Factor & has been published & validated for use in ELISA, WB, IH.
Immunohistochemistry: 5-10μg/mL (see suggested protocol).

Immunoblotting: 5-10μg/mL. BDNF is not easily extracted in standard buffers. Dissected tissues should be homogenized in 10 volumes of 100mM phosphate buffer containing 1mM EDTA, 2M guanidine hydrochloride (pH7.2), and three protease inhibitors, 10mM N-ethylmaleimide, 0.36mM pepstatin, and 1mM PMSF. Homogenates are then sonicated and centrifuged at 46K x g for 30 minutes at 4C. BDNF migrates as 18kDa, with a homodimer at ~27-30kDa. 1:1,000

ELISA: 5-10μg/mL

BIOLOGICAL ACTIVITY: Neutralizes BDNF, but not other neurotrophins.

Optimal working dilutions must be determined by end user.

Biochem/physiol Actions

Brain Derived Neurotrophic Factor (BDNF). By dot blot, less than 1% cross-reactivity against NGF, NT3 or NT4.

Physical form

Format: Purified
Immunoglobulin fraction lyophilized from PBS, with no preservatives. Reconstitute with 500 μL of sterile distilled water (1 mg/mL).

Preparation Note

Maintain lyophilized material at -20 to -70°C for up to one year after date of receipt. After reconstitution maintain at -20 to -70°C in undiluted aliquots for up to six months. Glycerol (ACS grade or better) can be added (1:1) for additional stability.

Brain-derived neurotrophic factor (BDNF) is involved in many aspects of neuronal biology and hippocampal physiology. Status epilepticus (SE) is a condition in which prolonged seizures lead to neuronal degeneration. SE-induced in rodents serves as a model of Temporal Lobe Epilepsy
BDNF overexpression in the ventral tegmental area prolongs social defeat stress-induced cross-sensitization to amphetamine and increases ?FosB expression in mesocorticolimbic regions of rats.

Stress is a major risk factor for substance abuse. Intermittent social defeat stress increases drug self-administration (SA) and elevates brain-derived neurotrophic factor (BDNF) expression in the ventral tegmental area (VTA) in rats. Intra-VTA BDNF overexpression enhances social defeat stress-induced cross-sensitization

Although it is well established that both follicular assembly and the initiation of follicle growth in the mammalian ovary occur independently of pituitary hormone support, the factors controlling these processes remain poorly understood. We now report that neurotrophins (NTs) signaling

Intermittent social defeat stress exposure augments behavioral response to psychostimulants in a process termed cross-sensitization. Brain-derived neurotrophic factor (BDNF) mediates synaptic plasticity and cellular responses to stress and drugs of abuse. We previously showed that repeated social defeat stress persistently
